A. As a rule mice should be primed with pristane only once; if it is necessary to repeat the pristane injection, one must wait 4 weeks after initial injection. In either case, one should wait one week after priming before injecting the hybridoma cells. A volum of 0.2 ml of pristane is recommended, with the maximum dose being 0.5 ml.
B. All ascites cell lines must be free of adventitious murine viruses. The number of hybridoma cells injected per animal should be within the range of 1 x 105 to 5 x 106 cells.
C. Animals must remain comfortable at all times. They should be routinely checked for normal breathing, ability to walk normally, and to readily reach food and water. Abdominal distension should not be allowed to progress to the point that creates sight of discomfort in the animals. If discomfort/impairment is detected, then the final fluid collection should be made promptly, under anesthesia, and the animal sacrificed.
D. Abdominal tapping (draining) must be performed under anesthesia. It should be limited to 3 times, with the third time being terminal.
